Objectives students should get an insight into molecularbiology and common techniques in the lab dealing with DNA cloning and PCR.

  • basics of DNA cloning
  • structure of DNA plasmids
  • PCR techniques in medical diagnostics and forensics
  • working with biological material and laboratory equipment
  • working under sterile conditions
Subject The course is devided into 3 parts:

1. in-frame cloning of a fusion gene via restriction enzymes

              reading of plasmid maps
              restriction enzymes
              DNA gel electrophoresis
              DNA isolation and ligation
              transformation of E.coli cells
              DNA preparation

2. POC - point of care analysis

              sample preparation
              PCR
              analysis

3. STR analysis

              sample preparation
              amplification of STR D1S80 via PCR
              DNA gel electrophoresis
              analysis of allelic frequency
