Ziele (*)Understanding of the fundamental principles of NMR spectroscopy, interpretation of spectra of simple bio-organic molecules, overview of the applications of NMR for biomolecular structure and interaction analysis.
Lehrinhalte (*)This course deals with the fundamental principles of nuclear magnetic resonance (NMR) spectroscopy, the interpretation of NMR spectra of simple organic and biological molecules, and gives a first overview of the applications of NMR for biomolecular structure and interaction analysis. In the section on fundamentals of Nuclear Magnetic Resonance Spectroscopy the main Chapter will be: (1) General Introduction to Spectroscopy including comparison to other techniques. (2) The physical foundations of NMR with special emphasis on liquid state 1H and 13C NMR. (3) The concept of shielding and the chemical shift for biological molecules. (4) The chapter on scalar Spin-Spin-Coupling is split into subchapters on homonuclear and heteronuclear couplings. The concept of decoupling is also introduced here. (5) Peculiarities of 13C NMR at natural abundance and with isotope enrichment are dealt with in this section. (6) The concept of nuclear spin relaxation is introduced with longitudinal and transverse relaxation. (7) The Nuclear Overhauser Effect is all important in biomolecular NMR and therefore is handled in a separate section. (8) Basic Two Dimensional NMR techniques are introduced with fundamental principles and applications. (9) NMR Structure Analysis is taught with special reference to biological molecules including macro-molecules. (10) Finally an outlook on industrial and medical applications of NMR spectroscopy
