(*)Students will be able to understand and describe the procedure of NMR resonance assignment of proteins using advanced multinuclear and multidimensional NMR experiments. Students can actively and independently work with protein 3D NMR spectra. Students understand how NMR spectroscopy is applied to probe protein structure, dynamics, and interaction with ligands.
Fertigkeiten Kenntnisse
(*)Students can describe the procedure of NMR resonance assignment by 3D triple resonance experiments like HNCO, HNCA, etc. (k2,k3). Students can work with 2D and 3D NMR data in specialized software programs and are able to perform a resonance assignment (k3,k4). Students understand advance NMR experiments with approaches for solvent suppression, and can decide which strategy is best applied in a certain context (k2,k5). Students understand how NMR can be applied to probe protein structure, dynamics, and interaction with ligands using advanced NMR spectroscopy methods (k2,k3). (*)- Advanced protein NMR techniques; pulsed field gradients, selective and non-selective RF pulses, solvent suppression strategies, relaxation editing, diffusion editing - Triple resonance techniques: HNCO, HNCA, etc. - Protocols for resonance assignment using triple resonance NMR and practical training - Protein NMR techniques for structural characterization e.g NOESY and other dipolar techniques) and for applications to protein dynamics (spin relaxation) and interactions (saturation transfer and editing techniques)
Beurteilungskriterien (*)Assignments, participation, presenting assignment results. Oral presentation of two review articles.
