Original study plan |
Bachelor's programme Biological Chemistry 2022W |
Objectives |
The biophysical laboratory aims at conveying students with basic knowledge in Biophysics.
|
Subject |
Students have to conduct four out of five offered experiments with a scope as diverse as: (i) Electrical characterization of a free standing planar lipid bilayers with a two electrode setup and measuring the single channel conductivity and life time of reconstituted gramicidin A. (ii) Estimating the critical micelle concentration of a detergent and its complexing stoichiometry with β-cyclodextrin with the help of contact angle measurements on water droplets. (iii) Measuring the binding affinity of a fluorescent probe to lipid vesicles and calculating the respective surface charge density and surface potential from it. (iv) Analyzing the membrane fusion of lipid vesicles via turbimetry, fluorescence resonance energy transfer and fluorescent self-quenching to be able to differentiate between aggregation, semi-fusion and fusion. (v) Estimating weak acid permeation across planar lipid bilayers from the transmembrane potential generated in the presence of a proton carrier.
